How much time do you have? I’ve recently spent six days in Berlin, seen a lot but still only scratched the surface. You could also stay for a week in Munich. It depends on your interests: shopping, sightseeing, museums, day trips outside the city etc. One or two days should be enough for Düsseldorf unless you want to go on daytrips to see other places in the area.
If you want to hike you could spend weeks in the Black Forest, and even if you rent a car and drive through you could stay there for a while. Again, it depends on your interests.

As EllyRigby says, it really depends on your interests. 1-2 days should be sufficent to give you the flavor of a place. However, almost anywhere in Germany you could spend weeks or months and not see all the excellent places to visit there or within 100 km (60 miles). You have mentioned four places that are not that close to each other, so you will spend some time traveling between them. If you only have a short vacation, you might want to limit the number of places you visit, and also consider places lying between them. For instance, the most scenc part of the Rhine lies between Duesseldorf and the Black Forest, between Koblenz and Ruedesheim.
