We will overnight in Weimar, and then head towards Hameln. We need to overnight someplace between Weimar and Hameln, possibly northeast or close to Kassel. What%26#39;s the most interesting town with good restaurants and nice old-city in that area?

You really can cover the Weimar-Hameln distance in one day (by car). But, if you have to choose, I definitely would pick Eisenach.
